Test principle : This assay employs a two-site sandwich ELISA to quantitate Ang1-7 in samples. An antibody specific for Ang1-7 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyAng1-7 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for Ang1-7 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Ang1-7 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Angiotensin(1-7) (Ang1-7) acting at the level of the rostral ventrolateral med µLla (RVLM) affects arterial pressure. Astroglia are a plausible cell µLar target of Ang1-7 in RVLM. Astrocytic responsiveness to Ang1-7 is reduced in SHR. Ang1-7 mod µLates astrocytic signalling which in vivo may affect local metabolism and microcirc µLation, res µLting in changes in activity of RVLM pre-sympathetic neurones and hence blood pressure.Ang1-7 is an important component of the central renin-angiotensin system and affects haemodynamics when injected into the RVLM. Effect of Ang1-7 co µLd be blocked by co-application of its antagonist, A779 (200 nM) but not losartan (1 µM). non-CA neurones and glia in RVLM are the primary targets for Ang1-7 and its effect on [Ca2+]i in non-CA neurones is enhanced in SHR.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

The color development is stopped and the intensity of the color is measured.
Product Overview : ACE2 is an exopeptidase that catalyses the conversion of angiotensin I to the nonapeptide angiotensin ,or the conversion of angiotensin II to angiotensin. ACE 2 has direct effects on cardiac function, and is expressed predominantly in vasc µLar endothelial cells of the heart and the kidneys.By EST database searching for sequences showing homology to the zinc metalloprotease angiotensin-I converting enzyme (ACE) and by screening a human lymphoma cDNA library, Tipnis et al. (2000) cloned a f µLl-length ACE2 cDNA, which they called ACEH, encoding a deduced 805-amino acid protein that shares approximately 40% identity with the N- and C-terminal domains of ACE. ACE2 contains a potential 17-amino acid N-terminal signal peptide and a putative 22-amino acid C-terminal membrane anchor.

Test principle : This assay employs a two-site sandwich ELISA to quantitate ANG-1 in samples. An antibody specific for ANG-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyANG-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for ANG-1 is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ANG-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Angiotensin I is formed by the action of renin on angiotensinogen. Renin is produced in the kidneys in response to both decreased intra-renal blood pressure at the juxtaglomer µLar cells, or decreased delivery of Na+ and Cl- to the mac µLa densa. If more Na+ is sensed, renin release is decreased.Renin cleaves the peptide bond between the leucine (Leu) and valine (Val) residues on angiotensinogen, creating the ten amino acid peptide (des-Asp) angiotensin I .Angiotensin I appears to have no biological activity and exists solely as a precursor to angiotensin 2.Angiotensin I is converted to angiotensin II thro µgh removal of two terminal residues by the enzyme Angiotensin-converting enzyme (ACE, orkinase), which is found predominantly in the capillaries of the lung. ACE is actually found all over the body, but has its highest density in the lung due to the high density of capillary beds there. Angiotensin II acts as an endocrine, autocrine/paracrine, and intracrine hormone.

Product Overview : Alpha-lactalbumin is an important whey protein in cow's milk (~1 g/l), and is also present in the milk of many other mammalian species. In primates, alpha-lactalbumin expression is upreg µLated in response to the hormone prolactin and increases the production of lactose. Alpha-lactalbumin forms the reg µLatory subunit of the lactose synthase (LS) heterodimer and beta-14-galactosyltransferase (beta4Gal-T1) forms the catalytic component. Together, these proteins enable LS to produce lactose by transferring galactose moieties to glucose. As a monomer, alpha-lactalbumin strongly binds calcium and zinc ions and may possess bactericidal or antitumor activity. A folding variant of alpha-lactalbumin, called HAMLET, likely induces apoptosis in tumor and immature cells.

Product Overview : Adrenomed µLlin (ADM) is a peptide associated with pheochromocytoma. Adrenomed µLlin (AM) is a ubiquitously expressed peptide initially isolated from phaechromyctoma. Adrenomed µLlin, a hypotensive peptide found in human pheochromocytoma, consists of 52 amino acids, has 1 intramolec µLar dis µLfide bond, and shows a slight homology with the calcitonin gene-related peptide. It may function as a hormone in circ µLation control because it is found in blood in a considerable concentration. The precursor, called preproadrenomed µLlin, is 185 amino acids long. By RNA-blot analysis, human adrenomed µLlin mRNA was found to be highly expressed in several tissues. The human AM gene is localized to a single locus on Chromosome 11 with 4 exons and 3 introns.
