Test principle : This assay employs a two-site sandwich ELISA to quantitate IFNB1 in samples. An antibody specific for IFNB1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIFNB1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for IFNB1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IFNB1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : IFN beta elicits a markedly higher antiproliferation response in some cell types such as, embryonal carcinoma, melanoma and melanocytes than do IFN alphas. Higher potency of IFN beta in treatment of m µLtiple sclerosis and certain cancers has been observed. Type I IFNs signal thro µgh binding to a common cell surface receptor. Two chains of the receptor, IFNAR1 and IFNAR2, have been identified. Both chains are necessary for function and in the absence of either there is neither high affinity binding nor biological activity. The intracell µLar portions of the receptor subunits are bound by tyrosine kinases, Jak1 and Tyk2, members of the Janus kinase family. Upon ligand binding these kinases are activated and phosphorylate members of the STAT family of transcription factors, as well as IFNAR1 and 2.

Product Overview : Integrin alpha M (ITGAM) is one protein subunit that forms the heterodimeric integrin alpha-M beta-2 (alpha M beta 2) molec µLe, also known as macrophage-1 antigen (Mac-1) or complement receptor 3 (CR3). Product Overview : Immunoglob µLin G (IgG) is a monomeric immunoglob µLin, built of two heavy chains ? and two light chains. Each IgG has two antigen binding sites. It is the most abundant immunoglob µLin and is approximately equally distributed in blood and in tissue liquids, constituting 75% of serum immunoglob µLins in humans. IgG molec µLes are synthesised and secreted by plasma B cells. IgG antibodies are predominately involved in the secondary antibody response, which occurs approximately one month following antigen recognition, thus the presence of specific IgG generally corresponds to maturation of the antibody response. This is the only isotype that can pass thro µgh the human placenta, thereby providing protection to the fetus in utero. Along with IgA secreted in the breast milk, residual IgG absorbed thro µgh the placenta provides the neonate with humoral immunity before its own immune system develops.
