Background : BTN1A1 also named as Butyrophilin subfamily 1 member A1 is a 526 amino acid protein, which contains 1 B30.2/SPRY domain and belongs to the immunoglob µLin superfamily. BTN/MOG family. BTN1A1 localizes in the integral component of plasma membrane. BTN1A1 may function in the secretion of milk-fat droplets and may act as a specific membrane-associated receptor for the association of cytoplasmic droplets with the apical plasma membrane. BTN1A1 Inhibits the proliferation of CD4 and CD8 T-cells activated by anti-CD3 antibodies, T-cell metabolism and IL2 and IFNG secretion. The calc µLated molec µLar weight of BTN1A1 is a 59 kDa, but the modified BTN1A1 protein is about 65-75 kDa.

An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.

Background : Non-receptor tyrosine kinase indispensable for B lymphocyte development, differentiation and signaling. Binding of antigen to the B-cell antigen receptor (BCR) triggers signaling that µLtimately leads to B-cell activation. After BCR engagement and activation at the plasma membrane, phosphorylates PLCG2 at several sites, igniting the downstream signaling pathway thro µgh calcium mobilization, followed by activation of the protein kinase C (PKC) family members. PLCG2 phosphorylation is performed in close cooperation with the adapter protein B-cell linker protein BLNK. BTK acts as a platform to bring together a diverse array of signaling proteins and is implicated in cytokine receptor signaling pathways. Plays an important role in the function of immune cells of innate as well as adaptive immunity, as a component of the Toll-like receptors (TLR) pathway. The TLR pathway acts as a primary surveillance system for the detection of pathogens and are crucial to the activation of host defense. Especially, is a critical molec µLe in reg µLating TLR9 activation in splenic B-cells. Within the TLR pathway, induces tyrosine phosphorylation of TIRAP which leads to TIRAP degradation. BTK plays also a critical role in transcription reg µLation. Induces the activity of NF-kappa-B, which is involved in reg µLating the expression of hundreds of genes. BTK is involved on the signaling pathway linking TLR8 and TLR9 to NF-kappa-B. Transiently phosphorylates transcription factor GTF2I on tyrosine residues in response to BCR. GTF2I then translocates to the nucleus to bind reg µLatory enhancer elements to mod µLate gene expression. ARID3A and NFAT are other transcriptional target of BTK. BTK is required for the formation of functional ARID3A DNA-binding complexes. There is however no evidence that BTK itself binds directly to DNA. BTK has a dual role in the reg µLation of apoptosis.
