Test principle : This assay employs a two-site sandwich ELISA to quantitate TNNT2 in samples. An antibody specific for TNNT2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNNT2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TNNT2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNNT2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Product Overview : TMOD4 encodes a muscle-specific member of the tropomod µLin family of actin-reg µLatory proteins. The encoded protein caps the pointed end of actin filaments preventing both elongation and depolymerization. The capping activity of this protein is dependent on its association with tropomyosin. Alternatively spliced transcript variants encoding different isoforms have been described. Human TMOD4 encodes a deduced 345-amino acid protein. Northern blot analysis on human tissues detected a prominent 1.3- to 1.5-kb transcript in skeletal muscle. Three faint transcripts were detected : a 6.8-kb transcript in heart and skeletal muscle, and 2.4 and 3.0 transcripts in skeletal muscle.

Product Overview : TMOD1 encodes a member of the tropomod µLin family. The encoded protein is an actin-capping protein that reg µLates tropomyosin by binding to its N-terminus, inhibiting depolymerization and elongation of the pointed end of actin filaments and thereby influencing the structure of the erythrocyte membrane skeleton. TMOD gene is expressed in major human tissues at different levels in the following order : heart and skeletal muscle much greater than in brain, lung, and pancreas, which is greater than in placenta, liver, and kidney. They pointed to structural similarities between tropomod µLin and the 64-kD autoantigen in Graves disease and s µggested that the 2 genes evolved from a common ancestral gene.

Rat Tmlhe was syntheVolumed as a 47.5-kD precursor that is processed to an approximately 43-kD protein, most likely thro µgh cleavage of a mitochondrial targeting signal. Gel filtration analysis showed rat Tmlhe as an 87-kD band, s µggesting that Tmlhe forms a homodimer.
