After removing any unbound substances, a biotin-conj µgated antibody specific for Ub is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Ub bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Ubiquitin is a protein encoded by the UBB gene.one of the most conserved proteins known. Ubiquitin is required for ATP-dependent, nonlysosomal intracell µLar protein degradation of abnormal proteins and normal proteins with a rapid turnover. Ubiquitin is covalently bound to proteins to be degraded, and presumably labels these proteins for degradation. Ubiquitin also binds to histone H2A in actively transcribed regions but does not cause histone H2A degradation, s µggesting that ubiquitin is also involved in reg µLation of gene expression. This gene consists of three direct repeats of the ubiquitin coding sequence with no spacer sequence. Consequently, the protein is expressed as a polyubiquitin precursor with a final amino acid after the last repeat. Product Overview : Troponin I is part of a heteromeric complex playing an important role in the reg µLation of skeletal and cardiac muscle contraction. It consists of three subunits, troponin I (TnI), troponin T (TnT) and troponin C (TnC). Each subunit is responsible for part of troponin complex function. TnI inhibits ATPase activity of acto myosin and TnT and TnI are present in cardiac muscles in different forms than in skeletal muscles. Only one tissue specific isoform of TnI is described for cardiac muscle tissue (cTnI) and this is expressed only in myocardium.Equally reactive with free cardiac troponin I (cTnI) and cTnI forming complexes with other troponin components.
