Test principle : This assay employs a two-site sandwich ELISA to quantitate TNNT3 in samples. An antibody specific for TNNT3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNNT3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for TNNT3 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNNT3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Subunit T binds the troponin complex to the tropomyosin complex and is also required for Ca(2+)-mediated activation of actomyosin ATPase activity. There are 3 different troponin T genes that encode tissue-specific isoforms of subunit T for fast skeletal-, slow skeletal-, and cardiac-muscle. This gene encodes fast skeletal troponin T protein; also known as troponin T type 3. Alternative splicing res µLts in m µLtiple transcript variants encoding additional distinct troponin T type 3 isoforms. A developmentally reg µLated switch between fetal/neonatal and ad µLt troponin T type 3 isoforms occurs. Additional splice variants have been described but their biological validity has not been established. Mutations in this gene may cause distal arthrogryposis m µLtiplex congenita type 2B (DA2B).

Product Overview : TNNI2 is a fast-twitch skeletal muscle protein, a member of the troponin I gene family, and a component of the troponin complex including troponin T, troponin C and troponin I subunits. The troponin complex, along with tropomyosin, is responsible for the calcium-dependent reg µLation of striated muscle contraction. Mouse studies show that this component is also present in vasc µLar smooth muscle and may play a role in reg µLation of smooth muscle function. In addition to muscle tissues, this protein is found in corneal epithelium, cartilage where it is an inhibitor of angiogenesis to inhibit tumor growth and metastasis, and mammary gland where it functions as a co-activator of estrogen receptor-related receptor alpha. This protein also suppresses tumor growth in human ovarian carcinoma.

Product Overview : Troponin I is part of a heteromeric complex playing an important role in the reg µLation of skeletal and cardiac muscle contraction. It consists of three subunits, troponin I (TnI), troponin T (TnT) and troponin C (TnC). Each subunit is responsible for part of troponin complex function. TnI inhibits ATPase activity of acto myosin and TnT and TnI are present in cardiac muscles in different forms than in skeletal muscles. Only one tissue specific isoform of TnI is described for cardiac muscle tissue (cTnI) and this is expressed only in myocardium.Equally reactive with free cardiac troponin I (cTnI) and cTnI forming complexes with other troponin components. (In the presence of 5 mM EDTA). Antibodies are not affected by heparin, phosphorylation, oxidation and troponin complex formation. No cross-reaction with skeletal muscle troponin I.

Product Overview : The troponin C protein is the calcium-binding subunit of the troponin complex. It occurs in 2 distinct isoforms : fast skeletal troponin C, which is expressed exclusively in fast-twitch skeletal muscle, and slow/cardiac troponin C, which is expressed in both cardiac and slow-twitch skeletal muscle.
