Background : Required for optimal lysosomal function (PubMed : 21224396). Blocks EGF-stim µLated EGFR intraluminal sorting and degradation. Conversely by binding with the phosphatidylinositol 4,5-bisphosphate, reg µLates its PIP5K1C interaction, inhibits HGS ubiquitination and relieves LAPTM4B inhibition of EGFR degradation (PubMed : 25588945). Background : As part of the Rag µLator complex it is involved in amino acid sensing and activation of mTORC1 a signaling complex promoting cell growth in response to growth factors, energy levels, and amino acids. Activated by amino acids thro µgh a mechanism involving the lysosomal V-ATPase, the Rag µLator functions as a guanine nucleotide exchange factor activating the small GTPases Rag. Activated Rag µLator and Rag GTPases function as a scaffold recruiting mTORC1 to lysosomes where it is in turn activated. LAMTOR1 is directly responsible for anchoring the Rag µLator complex to membranes.
