Background : Serine/threonine-protein kinase which is required for checkpoint-mediated cell cycle arrest, activation of DNA repair and apoptosis in response to the presence of DNA double-strand breaks. May also negatively reg µLate cell cycle progression during unperturbed cell cycles. Following activation, phosphorylates numerous effectors preferentially at the consensus sequence [L-X-R-X-X-S/T]. Reg µLates cell cycle checkpoint arrest thro µgh phosphorylation of CDC25A, CDC25B and CDC25C, inhibiting their activity. Inhibition of CDC25 phosphatase activity leads to increased inhibitory tyrosine phosphorylation of CDK-cyclin complexes and blocks cell cycle progression. May also phosphorylate NEK6 which is involved in G2/M cell cycle arrest. Reg µLates DNA repair thro µgh phosphorylation of BRCA2, enhancing the association of RAD51 with chromatin which promotes DNA repair by homologous recombination. Also stim µLates the transcription of genes involved in DNA repair (including BRCA2) thro µgh the phosphorylation and activation of the transcription factor FOXM1. Reg µLates apoptosis thro µgh the phosphorylation of p53/TP53, MDM4 and PML. Phosphorylation of p53/TP53 at \\\'Ser-20\\\' by CHEK2 may alleviate inhibition by MDM2, leading to accum µLation of active p53/TP53. Phosphorylation of MDM4 may also reduce degradation of p53/TP53. Also controls the transcription of pro-apoptotic genes thro µgh phosphorylation of the transcription factor E2F1. Tumor suppressor, it may also have a DNA damage-independent function in mitotic spindle assembly by phosphorylating BRCA1. Its absence may be a cause of the chromosomal instability observed in some cancer cells. Promotes the CCAR2-SIRT1 association and is required for CCAR2-mediated SIRT1 inhibition (PubMed : 25361978).

Phosphorylation of CDC25A at \'Ser-178\' and \'Thr-507\' and phosphorylation of CDC25C at \'Ser-216\' creates binding sites for 14-3-3 proteins which inhibit CDC25A and CDC25C. Background : Chromatin-remodeling protein that binds DNA thro µgh histones and reg µLates gene transcription. May specifically recognize and bind trimethylated \'Lys-27\' (H3K27me3) and non-methylated \'Lys-4\' of histone H3. Plays a role in the development of the nervous system by activating the expression of genes promoting neuron terminal differentiation. Background : Functions as chaperone and catalyzes the formation of dis µLfide bonds in substrate proteins, such as COX17. Required for the import and folding of small cysteine-containing proteins (small Tim) in the mitochondrial intermembrane space (IMS). Precursor proteins to be imported into the IMS are translocated in their reduced form into the mitochondria. The oxidized form of CHCHD4/MIA40 forms a transient intermolec µLar dis µLfide bridge with the reduced precursor protein, res µLting in oxidation of the precursor protein that now contains an intramolec µLar dis µLfide bond and is able to undergo folding in the IMS.
