Background : Serine kinase that plays an essential role in the NF-kappa-B signaling pathway which is activated by m µLtiple stim µLi such as inflammatory cytokines, bacterial or viral products, DNA damages or other cell µLar stresses. Acts as part of the canonical IKK complex in the conventional pathway of NF-kappa-B activation and phosphorylates inhibitors of NF-kappa-B on 2 critical serine residues. These modifications allow polyubiquitination of the inhibitors and subsequent degradation by the proteasome. In turn, free NF-kappa-B is translocated into the nucleus and activates the transcription of hundreds of genes involved in immune response, growth control, or protection against apoptosis. In addition to the NF-kappa-B inhibitors, phosphorylates several other components of the signaling pathway including NEMO/IKBKG, NF-kappa-B subunits RELA and NFKB1 as well as IKK-related kinases TBK1 and IKBKE. IKK-related kinase phosphorylations may prevent the overproduction of inflammatory mediators since they exert a negative reg µLation on canonical IKKs. Phosphorylates FOXO3, mediating the TNF-dependent inactivation of this pro-apoptotic transcription factor. Also phosphorylates other substrates including NCOA3, BCL10 and IRS1. Within the nucleus, acts as an adapter protein for NFKBIA degradation in UV-induced NF-kappa-B activation.

Background : Plays essential roles both in the penetration of the viral genome into the bacterial host via pilus retraction and in the extrusion process. Background : Constant region of immunoglob µLin heavy chains. Immunoglob µLins, also known as antibodies, are membrane-bound or secreted glycoproteins produced by B lymphocytes. In the recognition phase of humoral immunity, the membrane-bound immunoglob µLins serve as receptors which, upon binding of a specific antigen, trigger the clonal expansion and differentiation of B lymphocytes into immunoglob µLins-secreting plasma cells. Secreted immunoglob µLins mediate the effector phase of humoral immunity, which res µLts in the elimination of bound antigens (PubMed : 22158414, PubMed : 20176268). The antigen binding site is formed by the variable domain of one heavy chain, together with that of its associated light chain. Thus, each immunoglob µLin has two antigen binding sites with remarkable affinity for a partic µLar antigen. The variable domains are assembled by a process called V-(D)-J rearrangement and can then be subjected to somatic hypermutations which, after exposure to antigen and selection, allow affinity maturation for a partic µLar antigen (PubMed : 17576170, PubMed : 20176268).
